Is α-Casozepine Safe to Take Every Night?

The short answer
  • In published human trials lasting up to about 4 weeks per phase, α-casozepine was well tolerated. In the 2019 sleep trial, no one reported an adverse event while taking it.
  • It is not a hormone, and no study has reported dependence or withdrawal.
  • It is made from milk protein, so it is not for anyone with a milk-protein allergy.
  • Data beyond a few weeks per study are limited. If you are pregnant, nursing or take medication, ask your doctor first.

What the trials report

  • Kim et al., 2007: 63 women took 150 mg a day for 30 days and placebo for 30 days, with a 3-week break in between.
  • Kim et al., 2019: 48 adults took 300 mg a day for 4 weeks and placebo for 4 weeks. The only adverse event in the study, itching and hives, happened in the placebo phase; no one reported one while taking the hydrolysate. Blood tests and vital signs were also checked before and after each phase.
  • Chang et al., 2024: 36 adults took 600 mg a day for 2 weeks, then 300 mg for 2 weeks.
  • Thiagarajah et al., 2022: 4 weeks of the hydrolysate combined with L-theanine, reported without notable adverse events.

That is a clean record, but a short one: each trial phase lasted about a month. There are no published trials of nightly use over many months, and we would rather say so than imply otherwise.

Dose

Is it habit-forming?

α-casozepine is a food-derived peptide, not a drug and not a hormone. None of the trials reported dependence, and none reported withdrawal effects when people stopped, including in the crossover studies, where participants switched from the hydrolysate to placebo partway through. It is also not a sedative, so it is not meant to knock you out.

Who should not take it, or should ask first

  • Milk-protein allergy: do not take it. It is made from milk protein and it is not vegan.
  • Pregnant or nursing: it has not been studied in pregnancy or breastfeeding. Ask your doctor.
  • Taking medication, especially any prescription medicine for sleep or mood: ask your doctor before adding any supplement.
